In vitro production of RBCs from ES, iPS & Stem cell banking in Japan.

چکیده
The supply of transfusable red blood cells (RBCs) is not sufficient in many countries. If erythroid cell lines able to produce transfusable RBCs in vitro were established, they would be valuable resources. However, such cell lines have not been established. We developed a robust method to obtain differentiated cell lines following the induction of hematopoietic differentiation of mouse embryonic stem (ES) cells and established five independent hematopoietic cell lines using the method. Three of these lines exhibited characteristics of erythroid cells. Although their precise characteristics varied, each of these lines could differentiate in vitro into more mature erythroid cells, including enucleated RBCs. Following transplantation of these erythroid cells into mice suffering from acute anemia, the cells proliferated transiently, subsequently differentiated into functional RBCs, and significantly ameliorated the acute anemia.

I-54: New Models for Human and Mouse Genetic
The possibility to reprogram somatic human cells will greatly and deeply change genetic approach and allow the development of new tools to study genetics diseases. Indeed, our ability to study human genetic diseases suffers from the lack of valid in vitro models. The latter should (i) be originating from human primary cells, (ii) be able to self-renew for a long time and (iii) be able to differ...
متن کاملHuman Fetal Liver: An In Vitro Model of Erythropoiesis
We previously described the large-scale production of RBCs from hematopoietic stem cells (HSCs) of diverse sources. Our present efforts are focused to produce RBCs thanks to an unlimited source of stem cells. Human embryonic stem (ES) cells or induced pluripotent stem cell (iPS) are the natural candidates. Even if the proof of RBCs production from these sources has been done, their amplificatio...

Embryonic stem cells are pluripotent stem cells which have the ability to indefinitely self-renew and differentiate into all differentiated cells of the body. Regarding their two main properties (unlimited self-renewal and multi-lineage differentiation), these cells have various biomedical applications in basic research and cell based therapy. Because the transplantation of differentiated cells...
متن کاملEffects of Mouse Strain on Establishment of Embryonic Stem Cell Lines
Purpose: Embryonic stem (ES) cells are derived from the inner cell mass of blastocysts with self-renewal and pluripotency characteristics. These cells have potential for studies of in vitro differentiation, gene function, etc. This study was, therefore, initiated to establish new ES lines and evaluate the effects of strain on ES cell production. Materials and Methods: 3-5 day blastocysts were ...
متن کاملIn vitro Induction of Fetal Hemoglobin in Erythroid Cells Derived from CD133 Cells by Transforming Growth Factor-b and Stem Cell Factor
Increased fetal hemoglobin (HbF) in b-globin gene disorders ameliorates the clinical symptoms of the underlying disease. 5-azacytidine, butyrate and hydroxyurea, have been shown to activate g-globin gene expression. It has also been found that hematopoietic growth factors can influence expression of g-globin in erythroid cultures and in animal models. This study was designed to evaluate the in ...
